摘要
目的探讨Rho激酶抑制剂Y-27632对急性高眼压大鼠视网膜损伤的保护作用。方法 50只SD大鼠随机分为正常对照组(n=10)、生理盐水对照组(n=20,玻璃体腔内注射生理盐水)和Y-27632治疗组(n=20,玻璃体腔内注射100 nmolY-27632),选取右眼做为实验眼。正常对照组直接处死取材。生理盐水对照组及Y-27632治疗组分别用前房加压灌注法建立大鼠急性高眼压损伤模型,并于损伤后24和168 h取标本。HE染色观察视网膜组织病理学的改变,测量视网膜厚度;TUNEL染色检测细胞凋亡情况,计算凋亡指数;Western blot检测Caspase-3蛋白表达的变化。结果急性高眼压损伤24 h后,Y-27632治疗组视网膜凋亡细胞数量和Caspase-3蛋白表达水平均低于生理盐水对照组(P<0.01)。损伤168 h后Y-27632治疗组视网膜厚度大于生理盐水对照组(P<0.01)。结论 Y-27632可以减轻大鼠急性高眼压引起的视网膜损伤,具有神经保护作用。
Objective To investigate the neuroprotective effect of Rho kinase inhibitor Y-27632 on retinal injury after acute ocular hypertension in rats.Methods 50 Rats were randomly divided into normal control group,saline group(saline was injected into the vitreous) and Y-27632 group(100 nmol Y-27632 was injected into the vitreous).5 minutes after the injection,acute ocular hypertension was induced.The retinal tissues were collected 24 and 168 hours after injury for biochemical analysis,respectively.Histopathological changes of the tissues were observed by HE staining,and the thickness of retina was evaluated.Cell apoptosis was detected by TUNEL assay,and apoptotic index was calculated.Expression of Caspase-3 was detected by Western blot.Results Histological examination revealed that the retinal thickness was increased(P 〈 0.01) and fewer TUNEL-positive cells in the GCL and INL were found(P 〈 0.01) after injection of Y-27632.Caspase-3 expression was also inhibited significantly(P 〈 0.01).Conclusion The results suggest that the Rho kinase inhibitor Y-27632 can alleviate the retinal injury in rats with acute ocular hypertension and is therefore neuroprotective.
