摘要
目的:探讨肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)在大鼠慢性损伤骨骼肌中的表达及中药青白散对其的干预作用。方法:采用小鼠二甲苯性耳肿胀、大鼠蛋清性足肿胀和棉球性肉芽肿等多种炎症模型,观察青白散的抗炎作用;建立慢性软组织损伤动物模型,检测大鼠慢性软组织损伤后修复过程中TNF-α和IL-6水平的动态变化,并分析其相关性。结果:(1)青白散可明显抑制小鼠二甲苯性耳肿胀,显著抑制大鼠蛋清性足肿胀和棉球性肉芽组织增生。(2)模型组骨骼肌中TNF-α和IL-6的含量均较空白对照组显著增高(P<0.01)。(3)青白散组在不同时间点的骨骼肌中TNF-α和IL-6的含量均较模型对照组显著降低(P<0.01),与云南白药组比较差异无统计学意义。结论:青白散有明显的抗炎作用,能显著抑制大鼠慢性损伤骨骼肌中致炎因子TNF-α和IL-6的含量增高,该作用可能是青白散治疗慢性软组织损伤的主要机理之一。
Objective:To observe the dynamic changes in tissue TNF-αand IL-6 in chronic muscle injury model and effect of Qingbai powder.Methods:Animal models of inflammations including ear edema caused by xylene,plantar edema by albumen and granuloma by cotton ball in rats were made to observe the anti-inflammation effects of Qingbai powder.Chronic muscle injuring rat models were made and randomly divided into groups.QB Powder or Yunnan Baiyao were given to the model animals.The levels of muscle TNF-αand IL-6 were determined by the commercial Elisa kits.Results:QB Powder could not only significantly inhibit the ear edema in mice,but relieve the plantar edema and inhibit the granuloma in rats.During the skeletal muscle healing process,QB Powder could significantly decrease the advance of TNF-αand IL-6 in injured skeletal muscle(P〈0.01) and had no marked difference with that in the group treated with Yunnan Baiyao(P〉0.05).Conclusion:Qingbai powder has obvious anti-inflammation effects.It could inhabit the advancing level of TNF-αand IL-6 in injured skeletal muscle,this may be one of the mechanisms that Qingbai powder can treat chronic muscle injury.
