超量表达水稻miRNA 167a调控株型的研究 被引量:6

摘要 本研究将水稻MIR167基因家族成员之一MIR167a基因构建到玉米Ubiquitin基因组成型启动子下游,采用农杆菌介导的遗传转化方法实现了miRNA167a在水稻品种中花11号中的超量表达。Southern杂交进一步获得了单拷贝基因插入的转基因植株。Ubi::MIR167a超量表达植株表现为矮化、分蘖角度增大、有效分蘖数减少的株型变化,进一步分子检测表明突变表型与miRNA167a表达量呈共分离。通过生物信息学预测手段及表达量分析,初步确定ARF8为miRNA167a直接靶基因。本研究初步解释miRNA167a在调控水稻株型尤其是分蘖角度方面有重要功能。 To get the miRNA167a overexpressing transgenic rice plants,we constructed miRNA167a,one member of the MIR167 gene family,under the maize Ubiquitin promoter and transformed the vector into Zhonghua 11 by Agrobacterium-mediated transformation.We identified the single copy transgenic plant by Southern blotting.The miRNA167a overexpressing plants performed varieties of plant architecture changes,such as dwarf,increase in the till angles and decrease in the effective tiller numbers.Further molecular analyses showed the cosegregation between the phenotypes and the expression level of miRNA167a.ARF8 was identified as the target gene of miRNA167a in rice by bioinformatics prediction and gene expressing level examination.This study showed the important function of miRNA167a in plant architecture,especially in regulating the tiller angle in rice.
