摘要
在pH为7.40的Tris-HCl缓冲体系中,采用荧光光谱技术研究了染料木素与牛血清白蛋白(BSA)的相互作用。不同温度(290K、300K、310K、315K)下测得的猝灭常数,证实染料木素对BSA的荧光猝灭为静态猝灭过程。根据热力学参数焓变(ΔH)小于零和熵变(ΔS)小于零(ΔH=-48.905kJ/mol,ΔS=-71.435kJ/mol),可以判断染料木素与BSA之间主要靠氢键和范德华力相结合。生成自由能变(ΔG)为负值,表明染料木素与BSA的作用过程属自发过程。应用同步荧光光谱技术考察了染料木素对BSA构象的影响。
The interaction between Genistein and bovine serum albumin(BSA) in Tris-HCl buffer(pH=7.4) was studied by fluorescence spectroscopy.The quenching constants obtained under different temperatures(290K,300K,310K and 315K) indicated that the quenching of Genistein on the fluorescence of BSA is a static quenching process.The thermodynamic parameters,enthalpy change(ΔH) and entropy change(ΔS),were both less than zero(ΔH=-48.905 kJ/mol and ΔS=-71.435 J/mol),so it could be judged that the binding between Genistein and BSA was mainly driven by Van de Walls force and hydrogen bond.The Gibbs free energy change(ΔG) was negative,showing that the interaction between Genistein and BSA was a spontaneous process.The effect of Genistein on the conformation of BSA was investigated by synchronous fluorescence spectroscopy.
