摘要
目的通过对帕金森病(Parkinson’s disease,PD)大鼠中枢神经系统一氧化氮合成酶(NOS)3种同工酶的活性变化的研究,探讨帕金森病发病的可能因素和机制。方法用6-羟多巴胺(6-OHDA)毁损单侧黑质建立大鼠向健侧旋转运动的帕金森病动物模型;采用分光光度法检测神经元型一氧化氮合成酶(nNOS)、内皮型一氧化氮合成酶(eNOS)、诱导型一氧化氮合成酶(iNOS)活性变化。结果与正常对照组相比,帕金森病组大鼠eNOS活性无明显变化;nNOS活性升高(P<0.05);iNOS活性显著升高(P<0.01)。结论具有神经毒性作用的nNOS、iNOS的升高,具有神经保护作用的eNOS活性未相应增加,可能是帕金森病发生发展的重要因素之一。
Objective To explore possible mechanism and pathogenic factors involved through studying changes in the activi-ty of 3 isoenzymes of nitric oxide synthetase (NOS) in the central nervous system of the rat with Parkinson' s disease (PD). Methods The rat model of PD was developed with the injection of 6-hydroxy dopamine (6-OHDA)into the striatum. Changes in the activity of neuronal nitric oxide synthetase (nNOS), endothelia nitric oxide synthetase (eNOS) and induced nitric oxide synthetase (iNOS)were monitored by spoctrophotometry. Results When compared with the animals in the normal group, it was shown that no significant chan- ges in the activity of eNOS could be detected for the animals in the experimental group with Parkinson' s disease, and the activity of nNOS and iNOS increased obviously (P 〈 0.05, P 〈 0.01 ). Conclusion Increased levels of neurotoxic nNOS and iNOS and the decreased level of neuroprotective eNOS on the other hand seemed to be one of the important pathogenic factors for the occurrence and de- velopment of Parkinson' s disease.
