摘要
目的研究大豆异黄酮及雌马酚的雌激素样作用在DMBA诱导的去势大鼠乳腺肿瘤发生中的作用。方法时间分辨荧光免疫分析法检测血清中雌二醇、雌三醇含量,Western-blot法和RT-PCR法检测乳腺组织中ERα、ERβ蛋白及基因表达。结果 SI、雌马酚各剂量组乳腺肿瘤发生率显著低于阳性对照组;SI高、中剂量(1000、500 mg/kg饲料)组和雌马酚高、中剂量组(400、200mg/kg饲料)雌二醇含量显著高于阳性对照组,雌三醇差别无统计学意义(P>0.05);SI和雌马酚各剂量组ERβ基因及蛋白表达显著高于阳性对照组(P<0.05);SI低剂量组ERα基因及蛋白表达显著低于阳性对照组(P<0.05)。结论 SI和雌马酚能够降低去势大鼠乳腺肿瘤发病率,其作用可能与改变内源性雌激素水平及乳腺组织ER表达有关。
Objective To explore the effect of estrogen-like effects of soy isoflavones and its metabolite equol on DMBA induced breast cancer.Method The levels of serum estradiol and estriol were analyzed by Time-Resolved Immunofluorometric Assay(TRFIA).The expression of ERα、ERβ protein and gene in breast tissue were analyzed by Western-blot and RT-PCR.Results Compared with the model group,the breast cancer incidence in all doses of SI and equol groups decreased significantly.The level of serum estradiol in middle and high dose SI and equol groups was significantly higher than the model group while no difference in the serum estriol.The expression of ERβ protein and gene in all doses of SI and equol groups were significantly higher than the model group while only the low dose SI group was significantly lower than the model group for ERα protein and gene expression.Conclusion SI and equol reduced the incidence of breast cancer in ovariectomized rats,and changed the levels of serum estradiol and estriol,indicating that SI and equol had estrogenic effect,which may be mediated by ER.
