摘要
目的探讨热淋清颗粒治疗自身免疫性慢性前列腺炎(CP)动物模型的疗效与作用机制。方法60只Wistar人鼠根据文献制造免疫性前列腺炎动物模型,随机分成4组,每组15只,分别作为热淋清高组、热淋清低组、模型组和正常组。热淋清高组和热淋清低组分别按2g/kg、1g/kg给予热淋清颗粒灌胃,qd;正常组、模型组给了等量生理盐水灌胃,qd;30d后水合氯醛腹腔注射麻醉,手术切取前列腺,HE染色鉴定大鼠成炎情况以及免疫组化检测CD4+、CD8+表达情况。结果正常组前列腺组织正常,前列腺结构完整,未见炎性细胞浸润,模型组大鼠前列腺可见轻重不同的炎症细胞浸润。热淋清高组绝大部分基底膜结构完整,前列腺上皮呈立方或柱状,无破坏及增生,间质有极少量淋巴细胞浸润和少量纤维组织增生;热淋清低组局部组织结构完整性遭到不同程度的破坏,部分腺腔细胞不均匀增生,间质水肿及纤维组织增生较为明显,中等量炎性细胞浸润,多为淋巴细胞及单核细胞。热淋清组随着剂量增加,前列腺组织CD4+表达上调、CD8+表达下调,与正常组比较,差异有统计学意义P〈0.05,CD4+/CD8+比例下降。结论热淋清颗粒对自身免疫性前列腺炎有显著的治疗作用,并侵前列腺组织中CD4+表达上调、CD8+表达下调从而明显减轻模型大鼠前列腺的炎症反应。
Objective To study the efficacy of relinqing- granules on treatment of rats with autoimmunity chronic prostatitis and its mechanism. Methods Rat model with autoimmune prostatitis were established according to the published literature. Sixty rats were randomly divided into 4 groups (n=15) such as the high thermal Lin group, the low thermal Lin group, model group, normal group. Rats in the high thermal Lin group and the low thermal Lin group were respectively treated with 2g, lg/kg orally thermal Lin particles, once a day. Rats in the normal group and the model group were given normal saline orally, once a day. By the intraperitoneal injection of chloral hydrate anesthesia, the prostate of rat was resected by surgery. Inflammation cells in prostate tissue were observed by HE staining and the expressions of CD4+, CD8+ in prostate tissue were detected by immunohistochemistry. Results Rats in normal groul~. had normal prostate tissue with structural integrity, and no inflammatory cell infiltration in prostate tissue. Rats in the model group had different grades inflammatory cell infiltration in prostate tissue. For rats in high thermal Lin Ching group, most of the basement membrane showed structural integrity and the prostate epithelium cells with no damage and proliferation were cubic or columnar, A few lymphocytes infiltration and small amount of fibrous tissue hyperplasia were observed in interstitial. For rats in low thermal Lin group, structural integrity of local prostate tissue was slightly destroyed. Some uneven proliferation of glandular cells, interstitial edema and fibrosis were obviously observed. Moderate inflammatory cell infiltration(mostly lymphocytes and monocytes) were observed. For rats in thermal Lin group, expressions of CD4+,CD8+ were upregulated with the dosage increased(P 〈0.05). Conclusion Heat Lin effect on autoimmune prostatitis. The inflammation state in rats with autoimmune prostat CD4+ and downregulation of CD8+.
