摘要
目的:研究美宝湿润烧伤膏(MEBO)对大鼠深Ⅱ度烫伤创面愈合过程中内毒素(LPS)及Toll样受体4(TLR4)含量的影响。方法:30只SD大鼠,随机均分为空白对照、模型对照和MEBO组。复制大鼠深Ⅱ度烫伤创面模型,每天观察烫伤大鼠创面,记录创面愈合时间并计算创面愈合率。于烫伤后第1、3、7、14、21、28天分别通过尾静脉采血收集大鼠静脉血,分离血清、血浆。采用鲎试剂检测血浆中脂多糖(LPS)含量;采用酶联免疫吸附法检测血清中相关致炎细胞因子肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的水平;采用流式细胞术检测不同时相点大鼠外周血中TLR4阳性表达率。结果:与模型对照组比较,MEBO组大鼠创面愈合时间显著缩短(P<0.01);用MEBO7d后,大鼠血清中LPS、TNF-α、IL-6水平显著降低(P<0.01或P<0.05),大鼠外周血中TLR4阳性表达率显著降低(P<0.01或P<0.05)。结论:MEBO可促进大鼠深Ⅱ度烫伤创面愈合,其作用机制可能与降低烫伤大鼠LPS、TLR4以及相关的致炎细胞因子的表达有关。
OBJECTIVE: To study the effects of Meibao shirun shaoshang cream (MEBO) on the protection in rats during deep second-degree bum wound healing. METHODS: 30 SD rats were randomly divided into blank control group, model control group and MEBO group. After the establishment of rat deep second-degree bum wound, we observed the wound everyday and recorded the healing time to calculate healing rate of wound. Venous blood was collected at 3, 7, 14, 21 and 28 d from burn rats via vena candalis. Then, the serum and blood plasma were separated. Tachypleus amebocyte lysate (TAL) was used to assay the content of LPS in the plasma. Enzyme linked immunosorbent assay (ELISA) was employed to determine the levels of related proinflammatory cytokines such as TNF-α and IL-6 in the serum. Meanwhile, we used flow cytometry (FCM) to measure the positive percent of TLR4 in the peripheral blood at different time points, respectively. RESULTS: Compared with model control group, ME- BO could significantly shortened the healing time(P〈0.01). Meanwhile, MEBO decreased LPS, TNF-α and IL-6 levels in the serum at 7 d after burn (P〈0.01 or P〈0.05). FCM results suggested that MEBO obviously decreased the positive percent of TLR4 in the peripheral blood of burn rats at 7 d after bum(P〈0.01 or P〈0.05). CONCLUSION: MEBO could promote burn wound healing, which might.be associated with level reduction of LPS, TLR4 and related proinflammatory cytokines.
