摘要
目的以人重组TGF-β1蛋白作为抗原,制备兔抗人TGF-β1多克隆抗体。方法重组蛋白与弗氏佐剂等体积混匀,免疫新西兰白兔,获得兔抗人TGF-β1多克隆抗血清。饱和硫酸铵盐析法纯化多克隆抗体,间接酶联免疫吸附试验检测多克隆抗体效价,Western-blot技术进行抗体特异性检测。结果成功获得兔抗人TGF-β1多克隆抗体,纯化后抗体效价达1∶110 000。结论获得的兔抗人TGF-β1多克隆抗体具有良好的特异性,能满足进一步实验的需要。
Objective To prepare the rabbit-anti-human TGF-β1 polyclonal antibody from the immune rabbit with human TGF-β1 recombinant protein.Methods Equal volume of Freund′s adjuvant and recombinant protein were mixed,then New Zealand white rabbits were immunized with the the recombinant protein as antigen.The immune sera of rabbits were collected.Then antibody was purified by ammonium sulfate fractionation,then the polyclonal antibodies titer was detected by the indirect Enzyme-linked immunosorbent assay.Western-blot techniques was used to identify antibody specificity.Results The rabbit-anti-human TGF-β1 polyclonal antibody were successfully obtained.Purified rabbit anti-human polyclonal antibodies titer is 1∶ 10 000.Conclusion The specificity of rabbit anti-human TGF-β1 polyclonal antibodies is better to meet the need of further experiments.
