摘要
目的:观察红豆杉多糖对心肌缺血再灌注损伤(MIRI)导致的心肌细胞凋亡的影响,并初步探讨其可能机制。方法:将30只比格犬随机分为假手术组、模型组、红豆杉多糖低、高剂量组、卡维地洛对照组,每组6只,分别给药7d后,建立MIRI模型。采用TUNEL染色法标记缺血再灌注区凋亡心肌细胞,计数凋亡指数(AI);采用RT-PCR法及Western印迹法检测缺血再灌注区心肌Fas mRNA及蛋白表达。结果:模型组、低剂量组心肌细胞AI较假手术组显著增高(P<0.01);高剂量组、卡维地洛组心肌细胞AI显著低于模型组和低剂量组(P<0.05),与假手术组比较无显著差异;模型组及低剂量组Fas mRNA及蛋白表达较假手术组显著升高(P<0.01);各治疗组Fas mRNA及蛋白表达较模型组显著降低(P<0.01),且高剂量组、卡维地洛组Fas mRNA及蛋白表达显著低于低剂量组(P<0.05,P<0.01),与假手术组比较无显著差异。结论:抑制Fas mRNA及蛋白表达可能是红豆杉多糖抗缺血再灌注导致的心肌细胞凋亡的机制之一。
Objective: To investigate the influence of taxus polysaccharide on cardiomyocyte apoptosis in MIRI model of beagle dogs, and explore its possible molecular mechanism. Methods: 30 beagle dogs were randomly divided into sham operation group, model group, low-dose taxol polysaccharide group, high-dose taxol polysaccharide group, carvedilol group, 6 each group, and establish MIRI model after seven days administration. Marked myocardial cells of apoptosis in ischemia-reperfusion districts with TUNEL staining, counted apoptotic index (AI), detected Fas mRNA and protein expression of myocardial ischemia- reperfusion districts with RT-PCR method and western blotting. Results: AI of myocardial cells of the model group and low-dose group were higher significantly than the sham-operated group (P〈0.01); AI myocardial cells of the high-dose group and carvedilol group were lower significantly than the model group and low-dose group (P〈0.05), compared with the sham group no significant difference. Fas mRNA and protein expression of the model and low-dose group were higher significantly than the sham-operated group (P〈0.01); Fas mRNA and protein expression of the treatment group were lower significantly than the model group (P〈0.01), and protein expression of Fas mRNA of the high-dose and carvedilol group were lower significantly than the low-dose group (P〈0.05), compared with the sham group no significant difference. Conclusion: Taxus polysaccharides can inhibit myocardial apoptosis cause by ischemia-reperfusion, its mechanism may be related to reduced myocardial Fas mRNA and protein expression.
