丹参注射液对心肌梗死后大鼠缺血心肌血管新生的影响 被引量:24

Effect of Salvia Miltiorrhiza Injection on Angiogenesis in Ischemic Myocardium of Myocardial Infarction Rats

摘要 目的观察丹参注射液对心肌梗死后大鼠缺血心肌血管新生的作用及机制。方法选取Wistar大鼠32只结扎大鼠左冠状动脉,造成急性心肌梗死动物模型,随机分为丹参注射液组、麝香保心丸组、模型组,并设假手术组,每组8只。6周后检测各组大鼠缺血心肌中微血管数(MVC)、微血管密度(MVD)及VEGF mRNA的表达及其灰度值。结果模型组、麝香保心丸组及丹参注射液组大鼠心肌梗死边缘区MVC和MVD较假手术组明显增多(P<0.05);丹参注射液组、麝香保心丸组大鼠心肌梗死边缘区MVC和MVD较模型组明显增加(P<0.05),丹参注射液组与麝香保心丸组作用相似(P>0.05)。模型组VEGFmRNA表达及其灰度值明显高于假手术组(P<0.05);丹参注射液组、麝香保心丸组明显高于模型组(P<0.05),丹参注射液组低于麝香保心丸组(P<0.05)。结论丹参注射液可促进心肌梗死后大鼠缺血心肌血管新生,其机制可能与促进VEGFmRNA的表达有关。 Objective To observe the effect and mechanism of Danshen Injection (Salvia Miltiorrhiza Injection) on the an giogenesis in ischemic myocardium of the rats with myocardial infarction (MI). Methods The left coronary arteries of Wistar rats were ligated to make the models of acute myocardial infarction. Totally 32 rats were randomized into Salvia Miltiorrhiza Injection group, Shexiang Baoxin Pills (Pills containing Mosehus to protect the heart) group (positive control group), and model group (negative control group). A sham operation group was set up. Eight rats were included in each group. Microvascular count (MVC), Microvascular density (MVD), and vascular endothelial growth factor (VEGF) mRNA expression in ischemic myocardium of rats with MI were detected six weeks later. Results The MVC and MVD in the marginal zone of ischemic my ocardium of rats with MI in the model group, Shexiang Badxin Pills group, and Salvia Miltiorrhiza Injection group were significantly higher than those in the sham operation group (P〈0.05). The MVC and MVD in the marginal zone in the Salvia Milti- orrhiza Injection group and Shexiang Baoxin Pills group were significantly higher than those in the model group (P〈0.05). The effect of Salvia Miltiorrhiza Injection group was similar to that of Shexiang Baoxin Pills group (P〈0.05). The VEGF mRNA expression and its grayness in the model group was significantly higher than those in the sham operation group (P〈0.05), they were significantly higher in the Salvia Miltiorrhiza Injection group and Shexiang Baoxin Pills group than in the model group (P〈0.05), whilst they were lower in the Salvia Miltiorrhiza Injection group than in the Shexiang Baoxin Pills group (P〈 0.05). Conclusion Salvia Miltiorrhiza Injection may promote angiogenesis in ischemic myocardium of the rats with MI. Its mechanism might be related to promoting the VEGF mRNA expression.
