摘要
为了探索不同品种、不同外植体和不同激素配比对月季愈伤组织诱导的影响,优化其愈伤组织继代培养的培养基组成,以期为将来采用基因工程改良其遗传性状奠定理论基础,以贵阳市场上畅销的淑女、艳粉和卡罗拉等3个月季品种为试验材料,以MS为基本培养基分别进行了相关研究。结果表明:艳粉愈伤诱导率最高,淑女次之,卡罗拉较低,其诱导率分别为58.7%、57.9%和52.8%;最佳诱导条件:以花瓣为外植体在MS+NAA 4mg/L+6-BA 2mg/L培养基上进行培养较为理想;而愈伤组织继代培养在MS+6-BA 2mg/L+NAA 0.5mg/L培养基上进行培养较为理想;光照和黑暗对保持愈伤组织无显著影响。
The different explants of three R.chinensis varieties(Yanfen,Shunu and Carnola) were cultured on MS media with different hormone proportion to study the effects on calli induction of R.chinensis for optimizing medium composition of calli subculture.The results showed that the calli induction rates of Yanfen,Shunu and Carnola were 58.7%、57.9% and 52.8% respectively,the optimum explant was petal,the optimum media for calli induction and calli subculture were MS+NAA 4 mg/L+6-BA 2 mg/L and MS+6-BA 2.0 mg/L+NAA 0.5 mg/L separately,and there was no significant difference between light and dark condition for calli conservation.The research results provided the scientific basis for character modification of R.chinensis by genetic engineering in future.
