摘要
目的建立同时测定豚鼠血浆、脑组织、脑脊液和内耳外淋巴液中丹酚酸B和丹参酮IIA的高效液相色谱法。方法采用C18色谱柱,流动相2%冰醋酸水溶液-甲醇,梯度洗脱,流速1 ml.min-1,检测波长280 nm。结果该法能将丹酚酸B和丹参酮IIA有效分离。不同组织中药物在检测范围内线性良好,r均大于0.999,萃取回收率在74.76%~104.27%,方法回收率在93.82%~108.94%,日内和日间精密度RSD小于7.87%。结论该方法简单、灵敏,可用于豚鼠体内丹酚酸B和丹参酮IIA的测定,为二者体内过程及药动学研究奠定基础。
ObjectiveTo develop and validate an HPLC method for the simultaneous determination of Salvianolic acid B and Tanshinone IIA in plasma,brain tissue,cerebrospinal fluid and perilymph of guinea pig. MethodsChromatographic separation was performed on a C18 HPLC column,with gradient elution of 2% glacial acetic acid and methanol as mobile phase.The flow rate was 1 ml·min-1 and UV detector was set at 280nm. ResultsThe HPLC method showed good chromatographic resolution for Salvianolic acid B and Tanshinone IIA.The calibration curves were linear over the investigated concentration ranges with all correlation coefficients higher than 0.999.The extraction recoveries for the two components were between 74.76% and 104.27%,and the method recoveries were between 93.82% and 108.94%.The intra-and inter-day RSD were less than 7.87%. ConclusionThe proposed method is simple,selective and suitable for in vivo determination of Salvianolic acid B and Tanshinone IIA in guinea pig,which may provide the analytical method to their action mechanism and pharmacokinetics.
