摘要
目的探讨龙眼肉水提物对东莨菪碱所致学习记忆获得性障碍大鼠学习记忆能力的影响及其可能的作用机制。方法将50只雄性大鼠随机分5组,连续灌胃21 d。第16天开始进行Morris水迷宫行为学测试,测试前20 min,除空白组ip生理盐水(NS)外,其余各组均ip东莨菪碱溶液,第21天采集血清、海马和脑皮质标本,检测各组各标本中抗氧化的能力相关指标。结果与空白组比较,模型组有明显的学习记忆障碍,各标本的超氧化物歧化酶(SOD)和谷胱甘肽性氧化物歧化酶(GSH-Px)活性显著下降,丙二醛(MDA)含量明显升高(P<0.01);与模型组比较,龙眼肉水提物各组均能明显改善痴呆大鼠的学习记忆障碍,显著提高各标本SOD和GSH-Px活性,明显降低MDA含量(P<0.05或P<0.01)。结论龙眼肉水提物对东莨菪碱所致大鼠学习记忆获得性障碍有明显的改善作用,其作用机制可能与其抗氧化作用密切相关。
ObjectiveTo explore the effects of Aqueous extract of Arillus Longan(ALA) on learning and memory ability in scopolamine-induced dementia rats and its possible mechanism. MethodsFifty male rats were randomly divided into five groups and ALA given by P.O.for consecutive 21 days.Learning and memory ability was continually tested by Morris water maze on the 16th day.Drugs were intraperitoneally given 20 minutes before the test for rats in each group,normal saline in normal control group,scopolamine 3 mg·kg-1 in other groups.All rats were sacrificed to obtain the serum,hippocampus and cerebral cortex on the 21st day.The activities of SOD,GSH-Px and the content of MDA in serum,hippocampus and cerebral cortex were investigated. ResultsCompared with the dementia model control group,ALA obviously improved the learning and memory ability in scopolamine-induced rats,significantly increased the activities of SOD,GSH-Px and decreased the content of MDA in serum,hippocampus and cerebral cortex. ConclusionALA can significantly improve the learning and memory ability in learning and memory disorder rats induced by scopolamine,and its relevant mechanism may be closely related to its antioxidant effect.
