早期实验性近视眼视网膜色素上皮细胞的培养及超微结构变化的研究 被引量:2

摘要 目的:研究豚鼠早期实验性近视眼视网膜色素上皮(RPE)细胞的培养及超微结构的改变,探索近视的发病机制。方法:2周龄豚鼠30只随机分为A、B、C组,每组10只,再随机选取5只(10眼)正常2周龄豚鼠不作任何干预,作为正常对照眼。选取任意眼戴一10 DS凹透镜,分别饲养6 d、15d、30 d后除去镜片,验光及测眼轴长度确定近视形成后,采用细胞酶消化法培养豚鼠的前部及后部RPE细胞,取3~6代RPE细胞进行光、电镜检查。结果:前部及后极部RPE细胞分别于诱导15 d、6 d后出现内质网扩张、内质网及核糖体等细胞器均减少等改变。结论:豚鼠前部RPE细胞于诱导15 d后,细胞增殖减少;后极部RPE细胞于诱导6 d后,细胞增殖减少。 objective: To analyze the biological characteristics of guinea pig anterior and posterior retinal pigment epithelial cells.Methods: 30 two-week-old guinea pigs which were divided in three groups randomly were lens-induced monocularly,A spherical lenses of-10.00D were wear to unilateral eyes,another eye in each animal was untreated as self-control(SC).5 two-week-old guinea pigs were randomly closed as normal-control(NC).Each group was raised for 6、15、30 days separately,Before and after experiment,refraction was measured using retinoscopy and the oculus uterque axis oculi length determined by A-scan ultrasonography.The anterior and posterior retinal pigment epithelial cells of each group were cultured and passaged 3-6 generations in vitro.The cultured cells were studiedby light microscope and electron microscope.Results:The endoplasmic reticulum dilation and Organelles such as endoplasmic reticulum and Ribosome decrease of anterior and posterior retinal pigment epithelial cells were observed after 15 and 6 days induced.Conclusins:Cell proliferation of the anterior retinal pigment epithelial cells is reduced after 15 days induced,the same result happens in the posterior retinal pigment epithelial cells after 6 days induced.
