凝血酶对脊髓组织毒性损伤机制及水蛭素的干预作用 被引量:2

摘要 目的:研究凝血酶对大鼠脊髓组织毒性损伤机制及水蛭素的干预作用。方法:用免疫组化法检测凝血酶受体1(PAR-1)在脊髓组织中的表达,显微镜和MOTAC病理图像分析仪检测PAR-1免疫组化阳性细胞数;HE染色病理切片观察脊髓组织形态学的改变。结果:实验各组PAR-1阳性表达及脊髓组织水肿在脊髓损伤后6小时开始,72小时达到高峰(P<0.01);水蛭素可明显降低各时相PAR-1阳性表达(P<0.01或P<0.05)。结论:凝血酶对脊髓组织有细胞毒性损伤作用,且于72小时时损伤最严重;凝血酶抑制剂水蛭素可以选择性和特异性抑制凝血酶对脊髓组织的损伤。 Objective: To study mechanism of thrombin on spinal cord tissues toxic injury and intervention of himdin.Method:The PAR-1 expression of spinal cord tissues was detected by immunohistochemical method, and the count of PAR-1 immunohistochemical positive cell was tested by microscope and MOTAC pathological image analyzer. The morphology changes of spinal cord tissues were observed depend on HE dyed pathological slices. Result: The PAR-1 positive expression and spinal cord edema emerged in 6th hours after spinal cord injury and the peak at 72th hours (P〈0.01). Leeches element can obviously reduce the time phase PAR - 1 positive expression (P〈0.01 or P〈0.05).Conclusion:Thrombin had effect on cytotoxicity damage for spinal cord tissues, and the most is at 72th hours. Hirudin of thrombin inhibitor can selectively and specifically inhibit spinal cord injury of thrombin.
