原始巨核白血病细胞系(HI-Meg)诱导前后sis和mye癌基因表达的研究 被引量:1

摘要 我们对巨核细胞分化与癌基因表达的关系进行了初步研究。HI-Meg为原始巨核白血病细胞系,经维甲酸诱导后,该细胞向成熟巨核细胞方向分化。我们检测了C-myc及C-sis两种癌基因在诱导前后的HI-Meg细胞中mRNA表达的改变。结果表明,在诱导后的巨核细胞中,sis表达下降3.82倍,myc表达下降4.54倍,但这两个癌基因表达仍高于在正常淋巴细胞中的水平。本文讨论了这一结果对研究巨核细胞分化与调控及白血病发生的意义。 The relationship beween differentiation and expression of oncogenes on megakaryoblastic leukemia cells was studied. Hi-Meg is a human megakaryoblastic leukemia cell line established in our laboratory. Induced by RA, the cells would differentiate into promegakaryocytes. We have examined the expression of two kinds of oncogenes sis and myc on the induced and uninduced cells. The results showed that, after induced by RA, the cells revealed much less expression of oncogenes than uninduced cells, decreasing 3.82 times for sis gene, and 4.54 times for myc gene. However, the level of the expression in the induced Hi-Meg cells was still higher than that in the nomal lymphocytes. The significance for regulation of megakaryocytic differentiation and leukemogenesis is discussed.
