摘要
以小鼠迟发超敏反应(DTH)为指标,筛选了31种中药,发现其中大黄等七味中药水提物30g/kg×7d,口服给药能明显抑制绵羊红细胞(SRBC)所致的昆明种小鼠的DTH反应。L8(2′)正交实验结果表明,大黄等四味中药合用对DTH反应的抑制作用较其它配伍法为优;又L9(3′)正交实验结果表明,此四味中药等量配伍,作用最好。 对该方进行多种免疫指标的实验研究,结果如下:(1)该组方30、15、7.5g/kg×7d均能显著抑制NIH小鼠的DTH反应,P<0.01,但作用较地塞米松25mg/kg×2d和环磷酰胺25mg/kg×2d(给药量下同)为弱。(2)该组方30g/kg×7d对NIH小鼠T淋巴细胞转化有明显的抑制作用。(3)该组方30g/kg×7d能明显抑制NIH小鼠对SRBC所致的凝集素抗体产生,P<0.01,但作用较地塞米松和环磷酰胺为弱,15g/kg×7d剂量无明显的抑制作用。(4)该组方对抗体形成细胞的抑制作用与凝集素相似, 30g/kg×7d组与环磷酰胺之间无明显差异。(5)该组方30、 15g/kg×7d能明显抑制小鼠单核吞噬系统对刚果红的廓清,P<0.01,而7.5g/kg×7d及Dex、CY 25mg/kg×2d对此无明显影响。(6)该组方30g/kg×7d能明显抑制小鼠腹腔巨噬细胞吞噬功能,P<0.01,地塞米松对此也有一定的抑制作用,P<0.05,而环磷酰胺虽有抑制趋势,但无统计学意义,三种药之间也无明显的统计学差异。(7)?
MYYZF is composed of Rhubarb and the other three herbs. Our research showed that the inhibitory effect was the greatest when the four herbs were with same dosage on mice. The immunosuppressive effects of MYYZF, Dex and Cy on mice were compared.
The results showed that: The water extract of the formulation of MYYZF given orally to mice at a dose of 30g/kg for seven successive days, could inhibit murine DTH reaction induced by SRBC, decrease serum IgG and hemolysin produced by spleenocytes, decrease clearance rate of iv Congo red, suppress the phago-cytic function of peritoneal macrophages, suppress the proliferative response of spleenic lymphocytes induced by ConA, and decrease spleen and thymus weights. The experiment also showed that it was with stronger inhibitory effect on macrophagocytic system in mice, while weaker inhibitory effect on cellular and humoral immunity than cyclophosphamide or dexamethasone at a dose of 25mg/kg by abdominal administration for two days.
The detailed immunosuppressive mechanism of action of the formulation remains to be studied.
