摘要
目的探讨应用高压氧治疗大鼠脑缺血再灌注损伤后,生长相关蛋白-43(GAP-43)在神经系统的表达。方法成年健康雄性SD大鼠32只,随机分为高压氧组(n=16)和高压空气组(n=16),每组再分为脑缺血再灌注模型组(n=8)和假手术组(n=8)。应用线栓法制备大鼠脑缺血再灌注动物模型,免疫组化法检测GAP-43在神经系统中的表达情况。结果应用高压氧治疗大鼠脑缺血再灌注损伤后,模型组GAP-43表达高于假手术组(P<0.05);但应用高压空气治疗后,模型组GAP-43表达略高于假手术组,差异无统计学意义(P>0.05)。高压氧治疗模型组GAP-43表达高于高压空气治疗模型组,差异有统计学意义(P<0.05)。结论应用高压氧治疗能够增强GAP-43的表达,有利于受损神经系统的结构修复和功能恢复。
Objective To investigate the effect of hyperbaric oxygen therapy on the expression of growth associated protein-43 ( GAP-43 ) in the nervous system after cerebral ischemia-reperfusion injury in rats. Methods 32 male adult Sprague-Dawley(SD) rats were randomly divided into hyperbaric oxygen group (n=16) and high pressure air group (n=16). Each group was subdivided into two subgroups: model group (n=8) and sham operation group (n=8). The animal model of cerebral ischemia-reperfusion was established in rats by suture method. Expression of GAP-43 in the nervous system was detected by immunohistochemistry. Results With the application of hyperbaric oxygen, the expression of GAP-43 in model group was higher than that in sham operation group (P〈0.05). But in high pressure air group there was no significant difference between two subgroups (P〉0.05). The expression of GAP-43 in model group of hyperbaric oxygen was higher than that in model group of high pressure air (P〈0.05). Conclusion Hyperbaric oxygen therapy can enhance expression of GAP-43 and repair the structure and function of the damaged nervous system.
