摘要
microRNA-21(miR-21)编码基因定位于TMEM49基因编码区域内(17q23.2),通过其自身的启动区域完成转录及polyA尾的聚合,最终变成成熟的miRNA。miR-21与多种疾病相关。miR-21可作为一种肿瘤RNA,其基因簇对多种肿瘤具有促进作用,在肿瘤的发生和发展中有类似癌基因作用,参与肿瘤血管形成,与肿瘤细胞的增殖、侵袭及抗药性密切相关。miR-21也在心脏疾病及肺纤维化疾病中起重要作用。此外,miR-21还可以调节免疫过程,对不同信号传导的靶蛋白起关键作用。随着对miR-21功能研究的不断深入,miR-21可能成为早期诊断和基因治疗新的分子靶标。
The gene coding for pre-miR-21 is located within the intronic region of the TMEM49 gene (17q23.2), but pri-miR-21 is transcribed independently through its own promoter regions and terminated with its own poly(A) tail. After transcription, pri-miR-21 is finally processed into mature miRNA, MiR-21 is correlated with various diseases. As an oncomiRNA, miR-2 l promotes many cancers' growing, It is involved in angiogenesis, cancer cell proliferatiorl, invasiveness and so on. MiR-21 also plays a crucial role in cardiac and pulmonary fibrosis. In addition, miR-21 can regulate immunological process and affect many target proteins in various signaling pathways. MiR-2 l may function as an attractive target for genetic and pharmacological modulation along with investigation.
