摘要
目的探讨提高冠脉结扎法大鼠心梗模型制作存活率方法。方法大鼠麻醉后,经口腔插管连通动物呼吸机,开胸结扎其冠状动脉。于术后4周麻醉处死大鼠,取出心脏,观察心脏外形及梗死瘢痕,并在进行HE染色后于显微镜下观察其病理变化。结果此法制作心梗模型的成功率为100%,术后动物存活率为96.7%。术后4周取出的心脏,左心室扩大变形,结扎后引起缺血区心室壁变薄凹陷,颜色较周围心肌浅,心腔内膜纤维化。结论本实验在原有冠脉结扎制作心梗模型的基础上,通过精细化、改良及简化步骤,提高了心梗模型的存活率,使模型更为稳定,降低了大鼠血管结扎后的死亡率。为心肌梗死的基础研究提供了可靠模型,减少了动物模型不稳定给实验带来的偏倚。
Objective To explore a better way for improving the survival rate of myocardial infarction model rats.Methods The myocardial infarction model was produced by ligating left anterior descending branch coronary artery through left thoracotomy after injection of narcotic into abdominal cavity and trachea intubation via mouth.The rats were killed by injecting overdose of narcotic at week 4 after operation.The heart was taken out to observe the shape,infarct size and the pathological morphology by HE staining.Results The successful rate of modeling was 100%,and 96.7% of rats survived after operation.After operation for 4 weeks,the left ventricle became larger,the ischemic region became thinner and the endocardial tissues of the survival rats were fibrotic.Conclusion This modified method could improve the survival rate of animal model,and be used to make a stable model for the basic research of myocardial infarction,and reduce the bias in the experiment due to the unstable animal models.
