平榛AGAMOUS基因的克隆与生物信息学分析 被引量:4

Cloning and Bioinformatic Analysis of AGAMOUS Gene in Hazelnut(Corylus heterophylla Fischer)

摘要 以野生平榛(Corylus heterophylla Fischer)为试材,采用RT-PCR方法从花芽中获得了一个平榛AGAMOUS基因cDNA,命名为ChAG,GenBank登录号为JN828811。序列分析结果表明,ChAG基因编码一个长度为726 bp,编码241个氨基酸的开放阅读框。氨基酸序列分析显示,该基因属于MADS家族AG亚家族。序列比对和系统进化分析表明,ChAG基因与欧榛的亲缘关系最近,相似性达99%。采用生物信息学手段对ChAG基因的保守结构域、疏水性和二级结构等进行分析。 a full-length cDNA sequence of AGAMOUS gene was obtained from floral buds of Corylus heterophylla Fischer using RT-PCR,named as ChAG gene(GenBank accession No.JN828811).Sequence analysis indicated that ChAG genecontains an opening reading frame(ORF) of 726 bp encoding a 241 predicted amino acids,belongs to AG subfamily of MADS family.Sequence alignment and phylogenetic analysis revealed that ChAG gene shared more than 99% homology with Corylus avellana.Conserved functional domain,hydrophobicity/hydrophilicity,secondary structure,and homology tree of ChAG protein were analyzed by using bioinformatics methods
