大鼠低氧低温睡眠剥夺后血清TNF-α和IL-6水平变化的研究 被引量:3

摘要 目的研究低氧低温睡眠剥夺条件下大鼠血清肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)浓度变化的意义。方法选用成年健康SD大鼠,随机分为5组,每组5只:正常对照组(CC组)、单纯睡眠剥夺组(SD_0组)、低氧低温环境下睡眠剥夺1d组(SD_1组)、睡眠剥夺4d组(SD_4组)、睡眠剥夺7d组(SD_7组)。分别检测血清TNF-α和IL-6的浓度。结果 (1)TNF-α水平:与CC组比较,SD_1组、SD_4组、SD_7组TNF-α水平均显著升高,差异有统计学意义(P<0.05,P<0.01);与SD_0组比较,SD_1组差异无统计学意义(P>0.05),而SD_4组、SD_7组水平显著升高,差异有统计学意义(P<0.01)。随低氧低温睡眠剥夺时间的延长,TNF-α水平呈现逐步升高趋势。(2)IL-6水平:与CC组比较,SD_1组、SD_4、SD_7组IL-6水平显著升高,差异有统计学意义(P<0.01);与SD_0组比较,SD_1组、SD_4组、SD_7组水平均显著升高,差异有统计学意义(P<0.01)。随低氧低温睡眠剥夺时间的延长,IL-6水平呈现先升高后下降的趋势。结论 (1)大鼠血清TNF-α和IL-6的表达水平与低氧低温睡眠剥夺的严重程度及持续时间有关。(2)低氧低温睡眠剥夺引起的血清TNF-α和IL-6的增高,短期可能对机体脑细胞具有应激性保护作用,而长期持续增高可能对神经元有损伤作用。 Objective To investigate the concentration of TNF-αand IL-6 in blood serum level in sleep deprivation model of SD rats under hypoxia and hypothermia conditions.Methods Healthy adult SD rats were randomly allocated into five groups including antitheses group(CC group),sleep deprivation group(SD_0) and sleep deprivation group at the 1st day,the 4th day and the 7th day(SD_1,SD_4,SD_7) under hypoxia and hypothermia conditions.ELISA were used to test the concentration of the blood serum of TNF-αand IL-6.Results(1)Compared with the CC group,the concentration of blood serum TNF-a in SD_1,SD_4 and SD_7 group rise significantly(P0.05 and P0.01);there was no significant different in TNF-αconcentration between SD_0 and SD_1 group(P0.05),while the TNF-αconcentration in SD_4 and SD_7 group rise significantly(P0.01).The serum TNF-αconcentration tends to rise as the sleep deprivation prolongs.(2) Compared with the CC group,the concentration of blood scrum IL-6 in SD_1,SD_4 and SD_7 group rise significantly(P0.01);compared with the SD_0 group,the concentration of blood serum IL-6 in SD_1,SD4 and SD_7 group rise significantly(P0.01).The concentration of blood serum IL-6 tends to rise first and then decline as the sleep deprivation prolongs.Conclusion(1)The expression of TNF-αand IL-6 in blood serum of the sleep deprivation SD rats were related with degree and duration of sleep deprivation.(2) The rise in TNF-αand IL-6 caused by hypoxia,hypothermia and sleep deprivation might protect the brain cells in the short run,but longterm continued increase of its level would like to injure the neuron.
