摘要
采用分光光度法,以邻硝基苯基荧光酮(O-NPF)为显色剂,研究了O-NPF与头孢唑啉(CFZ)络合物的可见光谱性质及测定条件。结果表明:CFZ在4.545~72.72μg/mL范围内遵循比尔定律,回归方程为:A=0.668ρ+0.5341,检出限1.53μg/mL,表观摩尔吸光系数为5.41×104L.mol-1.cm-1,相关系数为R2=0.9891,并探讨了其反应机理。方法已用于尿样及生物样品中CFZ含量的测定。
The visible spectral characterist ic and the reaction condition of the complex of o-nitro phenyl fluorone(O-NPF) and cefazolin(CFZ) were investigated by spectrophotometry with O-NPF as a developer.The results showed that the concentration of CFZ obeyed the Beer's law in the range of 4.545~72.72μg /mL and the regression equation was A=0.668ρ+0.5341 with the correlation coefficient r of 0.9891,the apparent molar absorptivity was 5.41×104 L·moL-1·cm-1and the detection limit was 1.53μg /mL.Further more,the reaction mechanism had been explored.The proposed method has been successfully applied to the determination of CFZ in urine and biological samples,and the results were satisfying.
