摘要
以玄参中哈巴俄苷含量为指标,本论文研究了水煎煮法、渗漉法、半仿生法及超声波法四种提取方法的提取效率。同时建立了一种哈巴俄苷的高效,可靠的检测方法。将粉碎后的玄参原料分别用以上四种方法进行前处理得到四种供试品溶液,然后采用高效液相(HPLC)检测法进行哈巴俄苷含量的检测,从中选取效率最高的提取方法,对样品进行再处理,分别检测其线性关系、精密度、稳定性、重复性、回收率。其结果是:超声波提取法效率最高;得线性回归方程:Y=2E+06x-278.17,r=0.9997,线性关系良好;精密度RSD为1.24%,稳定性RSD为1.6%,重复性RSD为1.78%,回收率在89%-100%,都比较好,符合统计学要求。
This dissertation was launched, intending to research the extraction methods which had the best extraction efficiency by detecting the content of harpagoside in Scrophularia ningpoensis. Those researched subjects included water extraction method, pereolation method, semi bionic method and ultrasonic method. In order to get the sample solution, the S. ningpoen.;is which had been smashed into powder had to he processed by the four methods mentioned before respectively, and then detect the content of harpagoside by HPLC, choose the best extraction method and process the sample by this means. And after that, examine its linear relationship, precision, stability, repeatability, recovery rate. After a series of experiments, results could be concluded as follows: the ultrasonic method was the ideal one, the linear equation: Y = 2E + 06x-278. 17,r=0.9997,the precision:RSD% = 1.6% ,the stability:RSD% = 1. 78% ,the repeatability:RSD% = 1.78% ,the recovery rate ranged from 89% - 100%. All the results showed that this method was a perfect extraction method.
