摘要
目的研究褪黑素(Mel)作用下,结直肠癌细胞系RKO细胞增殖和凋亡发生的变化。方法 MTT法检测Mel对RKO细胞增殖的影响,并同时观察不同浓度Mel作用下相应受体表达的变化,通过Hoechst染色观察细胞核染色结果,判断细胞凋亡情况,并同时用Western blot观察凋亡相关蛋白表达的变化。结果毫摩尔级的Mel可以抑制细胞增殖,膜受体MEL1AR随着浓度的增加表达上调,Hoechst染色显示Mel作用下细胞凋亡增加,抑制凋亡的相关蛋白,如Bcl-2和Bcl-XL的表达明显下降,而促进凋亡的相关蛋白,如Bax、Bak等表达量相应提高。结论 Mel通过受体MEL1AR发挥作用,抑制细胞的增殖,并促进细胞凋亡。
Objective To study the effect of melatonin (Mel) on the proliferation and apoptosis of colorectal cancer cell line RKO. Methods MTT assay was detected the proliferation of RKO cells treated with different concentrations of Mel. The receptor protein of Mel was also detected. Hoechst staining was used to observe the apoptosis. Western blot was detected the proteins associated in the apoptosis. Results Mel of pharmacological concentration (mmol/L) inhibited the proliferation and promoted the apoptosis of RKO. Expression of MEL1AR protein was in- creased. The anti-apoptosis proteins such as Bcl-2 and Bcl-XL was downregulated while the apoptosis proteins such as Bax and Bak was upregulated. Conclusion Mel can inhibit the proliferation and promotes the apoptosis of RKO cells by increase the MEL1AR receptor proteins.
