摘要
目的:建立大鼠子宫肌瘤模型,探讨TGF-β3和MMP11在子宫肌瘤的表达。方法:将20只大鼠随机分成对照组和模型组,对照组肌内注射生理盐水,模型组采用苯甲酸雌二醇和黄体酮负荷法建立大鼠子宫肌瘤模型,观察大鼠子宫大体形态;HE染色镜下观察大鼠子宫平滑肌病理改变,免疫组织化学和RT-PCR方法检测TGF-β3和MMP11的表达。结果:模型大鼠子宫呈暗红色,水肿现象明显;镜下肌层增厚,平滑肌排列紊乱,呈瘤样改变;大鼠子宫肌瘤组织中TGF-β3和MMP11的表达水平明显高于正常大鼠子宫组织,具有统计学意义(P<0.05)。结论:雌孕激素负荷法制作大鼠子宫肌瘤模型成功,TGF-β3和MMP11在大鼠子宫肌瘤中表达较高。
Objective: To establish an appropriate model of uterine leiomyoma for drug development and pathogenesis research,to investigate the relationship of transferring growth factor-β_3(TGF-β_3),and matrix metalloproteinase 11(MMP11) with uterine leiomyoma.Methods: Twenty female rats were randomly divided into two groups: control group(group C) and model group(group M).Group C was treated with physiological saline,while rats in group M were treated with estradiol benzoate and progesterone loading method to produce uterine leiomyoma model.Gross morphological features of rat uterus were observed,and pathological changes of uterine smooth muscle were studied with HE staining me-thod when uterine leiomyoma model was established.The expressions of TGF-beta_3 and MMP11 were detected with immunohistochemical staining and RT-PCR.Results: In group M,the uterine was kermesinus and obvious edema can be seen;The muscular layer became thicker and smooth muscle cell arrangement became disordered;The expression levels of TGF-beta_3 and MMP11 in group M were obviously higher than those in group C(P0.05).Conclusions: Uterine leiomyoma model is successfully established.TGF-beta_3 and MMP11 have a close relationship with uterine leiomyoma.
