摘要 目的研究17-β雌二醇对大鼠海马神经元树突生长的影响及血清饥饿情况下对神经元存活率的影响。方法体外培养3d的海马神经元通过加入浓度为1nmol/L和2nmol/L的17-雌二醇,统计比较神经元树突的数目和长度;体外培养1d的海马神经元,加入17-雌二醇,培养6~7d后血清饥饿48h后进行NSE染色及MTT检测,检测神经元形态及神经元存活率。结果 17-雌二醇可显著增加存活神经元树突的长度和数量;并能够有效地抑制血清饥饿引起的细胞减少,增加其存活率。结论 17-β雌二醇对大鼠海马神经元的生长发育及存活具有保护性作用。 Objective To study the effect of 17-β estradiol on the growth of rat hippocampal neurons dendrite as well as the rate of serum starvation survival of neurons.Methods Adding a concentration of 1nmol/L and 20nmol/L of 17-β estradiol to cultured hippocampal neurons in vitro and then cultured for 3 days,then comparing the number and lengths of neurons dendrite.Adding 17-β estradiol to cultured 1d neurons,then cultured for 6 to 7 days,NSE and MTT were stained after serum starvation for 48 hours to detect neuronal morphology and neural survival rates.Results 17-β estradiol could significantly increase the survival of neurons and the dendritic length.It could also suppress the cell decrease induced by serum starvation and increase their survival.Conclusion Estrogen can protect the growth and survival of hippocampal neurons.
