摘要
研究维甲酸(retinoicacid,RA)对视网膜色素上皮(retinalpigmentepithelium,RPE)细胞生长、增殖的影响。方法:通过细胞生长曲线的测定,观察10-6molLRA对RPE细胞生长的影响;通过3HTdR掺入和液闪测定10-5、10-6、10-7、10-8、10-9、10-10molLRA作用RPE细胞5天、7天时每分钟脉冲值(CPM),并计算抑制率。结果:RA作用的前3天内,对RPE细胞生长无明显影响;3天后RA对RPE细胞生长起抑制作用,这种抑制作用一直持续到第8天。终浓度为01%的无水乙醇与空白对照相比,经统计学处理,差别不显著(p>005)。在10-10~10-5molL范围内,RA对RPE细胞DNA合成有明显抑制作用(p<001)。结论:RA可抑制RPE细胞生长、增殖,且呈量效关系,RA的这种作用与抑制RPE细胞DNA合成有关。
To investigate the effect of retinoic acid (RA) on the growth、proliferation of cultured rabbit RPE cells.Methods:10 -6 mol/L RA was added to cultures of rabbit RPE cells and growth curves were measured with cell counting.10 -5 、10 -6 、10 -7 、10 -8 、10 -9 、10 -10 mol/ L RA were added to RPE cells respectively and proliferation rates of the cells were measured with 3 H thymidine incorporation and liquid scintillation techniques on day 5 and 7.Rusults:Cell growth was inhibited after 3 days of RA treatment.RA at a dosage ranging from 10 -9 ~10 -5 mol/L inhibited cellular proliferation effectively.Conclusion:RA had an dose dependent inhibition on rabbit RPE cell growth、proliferation and DNA synthesis,and it may be of potential use clinically.
