摘要
目的:利用体外培养的大鼠乳鼠心肌细胞,探讨葛根素(Puerarin,Pue)对高糖(highglucose,HG)诱导的心肌细胞因子和钙离子的影响。方法:新生大鼠心肌细胞培养,除对照组外分别给予HG(25 mmol/L)、HG(25 mmol/L)+Pue(10-3mol/L)、HG(25 mmol/L)+Pue(2×10-3mol/L),检测培养心肌细胞存活率、内皮素(endothelin,ET)和一氧化氮(nitric oxide,NO)浓度及细胞内钙离子的变化。结果:与对照组相比,HG会使培养心肌细胞存活率和NO浓度下降,ET水平增加,心肌细胞内钙离子浓度上升(P<0.01);Pue可以抑制由于HG诱导所出现的上述情况,且2×10-3mol/L Pue组的效果更好。结论:Pue能抑制HG引起的心肌细胞因子浓度改变和钙超载,此效应可能是葛根素减少高糖诱导心肌细胞损伤发生的保护机制之一。
Objective To study the effect of Puerarin (Pue) on related cytokines and calcium of cardiomyocytes induced by highglucose (HG) in neonatal rats. Methods Myocardial cells of neonatal rats were cultured in vitro. The cardiomyocytes were treated with HG (25 mmol/L) , HG(25 mmoL/L) + Pue(10^-3mol/L) , HG(25 mmol/L) + Pue(2 × 10^-3mol/L)except control group. The survival rate, the NO and ET levels, and spontaneous [ Ca^2+ ]i transient were investigated. Results Compared with control group, HG reduced the survival rate and NO levels, but enhanced the ET levels and spontaneous[ Ca^2 + ]i transient ( P 〈 0.01 ). Pue showed the function on reducing the previous mentioned indices induced by HG. The 2 × 10^-3 mol/L Pue group shows the better result. Conclusions Pue plays an effective role in intervening the change of related cytokines and the calcium overload, which indicates Pue exerts potential protective actions in the prevetion of cardiac cell injury.
