摘要
目的观察姜黄素(CMN)诱导HO-1高表达及其对动脉粥样硬化(AS)大鼠血清白细胞介素-6(IL-6)、C反应蛋白(CRP)含量的影响。方法以健康雄性Wistar大鼠34只,随机分为正常对照组(A组,8只)、模型组(B组,10只)、CMN组(C组,8只)及抑制组(D组,8只),B、C、D组同法复制AS模型,C组加用CMN,D组加用CMN及锌原卟啉。第14周末处死所有大鼠取降主动脉检测HO-1表达、分布及活力,经心脏取血检测IL-6、CRP。结果 C组较B组HO-1表达量及活力明显增强(P<0.05),IL-6、CRP含量显著降低(P<0.05);D组较C组HO-1表达量及活力明显下降(P<0.05),IL-6、CRP含量显著增高(P<0.05)。结论 CMN可诱导HO-1高表达并显著减轻炎症因子水平,锌原卟啉Ⅸ抑制HO-1高表达可使CMN抗炎作用显著下降。
Objective To explore whether curcumin(CMN) could induce the expression of heme oxygenase-1 (HO-1) in rat's aortic artery and the contents of C-reactive protein(CRP), interleukin-6 (IL-6) in atherosclerotic rats. Methods 34 male Wistar rats were randomly divided into 4 groups as control group(group A, n = 8), model group(group B, n = 10), treatment group (group C, n = 8) and suppression group ( group D, n = 8). Rate model with AS was established by same method in group B,C and D,group C were treated with CMN and group D treated both CMN and ZnPP IX at the same time. All of the rates were executed at the end of 14 weeks' treatment,detected the expression and distribution and its activity about HO-1 protein in the aortic artery, and the blood were drawn via heart from all rats in each groups to evaluate the contents of IL-6, CRP. Results The expression and activity about HO-1 in group C were increased significantly than that in group B(P〈0.05) ,the contents of IL-6 and CRP in group C were lower than that in group B(P(0.05);The expression and activity about HO-1 in group D were decreased significantly than that in group C(P〈0. 05), the contents of IL-6 and CRP in group D were higher than that in group c (P 〈0. 05). Conclusion CMN could induce HO-1 protein overexpression and increase its activity and decrease the degree of inflammation,however,this induction were blocked significantly by ZnPPIX.
