星点设计-效应面法优选凉血通瘀颗粒醇提工艺 被引量:6

Alcohol Extracting Process Optimization Based on Central Composite Design and Response Surface Methodology for Liangxue Tongyu Granule

摘要 目的优选凉血通瘀颗粒醇提最优工艺。方法采用星点设计-效应面法,以乙醇浓度、乙醇用量、提取时间为主要影响因素;芦荟大黄素、大黄酚、芍药苷等6种成分含量为评价指标,通过对各因素的多元线性回归及二项式拟合,采用效应面法和等高线叠加优选最佳提取工艺,并进行预测分析。结果确定凉血通瘀颗粒醇提的最佳提取工艺为加12倍75%乙醇,提取2次,每次90min。结论星点设计-效应面法优选凉血通瘀方醇提工艺,实际值与预测值吻合度高,预测性良好,方法简单易行,对工业生产有一定的意义。 OBJECTIVE To optimize the alcohol extraction process for Liangxue Tongyu granule by central composit design and response surface methodology.METHODS Central composite design and response surface methodology was used to optimize the alcohol extracting process.The main influential factors of extraction efficiency were ethanol concentration,reflux time,and solvent ratio.The conditions of the extraction were evaluated by the six compositions of Peoniflorin,Aloe-emodin,Rhein,Emodin,Chrysophanol and Rheochrysidin.Multiple linear regression and binomial fitting models were performed with the main influential factors.Response surface and superposition of contour line were used to optimize the process and to predict the response in the optimal region.RESULTS Analysis indicated the optimum conditions of the extracting were 75% ethanol,12 fold solvent,extracting for 2 times and for 1.5 h.CONCLUSION It shows that the optimum process for extracting is simple,more convenient and with an higher precision,which is significant to industry production.
