摘要
目的 研究异单叶大黄素 (isorhapotigenin ,Gn 1) ,白藜芦醇 (resveratrol,Gn 2 )和二苯乙烯低聚体(stilbeneoligopolymer,Gn 3)等 3种Gn类化合物对小鼠腹腔巨噬细胞产生肿瘤坏死因子α(TNFα)的影响。方法 用L92 9作为靶细胞的结晶紫染色法测定Gn类化合物对巨噬细胞经LPS或重组人白细胞介素 8(rhIL 8)产生TNFα 的影响。结果 Gn 1,Gn 2和Gn 3均能剂量依赖性地抑制巨噬细胞经LPS刺激产生的TNFα,Gn 1和Gn 3均可剂量依赖性地抑制巨噬细胞经rhIL 8刺激产生TNFα。结论 3种Gn类化合物可抑制小鼠腹腔巨噬细胞生成TNFα,可能是它们抗炎作用的机制之一。
AIM To investigate the effects of isorhapotigenin (Gn 1), resveratrol (Gn 2) and stilbene oligopolymer (Gn 3) on TNF α production by murine peritoneal macrophages stimulated with LPS or rhIL 8. METHODS Inhibition was used to show the inhibitory effect of three compounds on TNF α production by murine peritoneal macrophages measured with crystal violet staining assay of L929 cells. RESULTS Isorhapotigenin(10 -5 ~10 -4 mol·L -1 ), resveratrol(10 -5 ~10 -4 mol·L -1 ) and stilbene oligopolymer(10 -6 ~10 -4 mol·L -1 ) were shown to inhibit TNF α production by murine peritoneal macrophages stimulated with LPS in a dose dependent manner, IC 50 are 4 9×10 -5 mol·L -1 ,5 2×10 -5 mol·L -1 and 3 6×10 -5 mol·L -1 respectively. Isorhapotigenin(5×10 -6 ~10 -4 mol·L -1 ) and stilbene oligopolymer(10 -6 ~10 -4 mol·L -1 ) were also found to dose dependently inhibit TNF α production by murine peritoneal macrophages stimulated with rhIL 8, their IC 50 are 9 3×10 -5 mol·L -1 and 4 6×10 -6 mol·L -1 respectively. CONCLUSION The results suggest that the three Gn compounds can inhibit TNF α production by murine peritoneal macrophages, which is probably one of the mechanisms of the anti inflammatory effects of isorhapotigenin, resveratrol and stilbene oligopolymer.
