不同理化处理对苏云金芽胞杆菌质粒稳定性的影响 被引量:3

Plasmid Stability of Bacillus thuringiensis Influenced by Different Physical and Chemical Treatments

摘要 研究了用溴化乙锭诱变、升温和添加SDS等 3种处理对苏云金芽胞杆菌质粒稳定性的影响。结果表明 ,用 4.5 μg/ml的溴化乙锭处理野生菌株YBT 1 5 2 0和HD 5 6 7,对其携带ICPs基因的质粒的稳定性无影响。升温至 42℃时 ,野生菌株YBT 1 4 6 3和YBT 96 0 3的携带ICPs基因的质粒分别有 0 .41 %和 0 .37%发生丢失 ,一些小质粒 (<2kb)也发生丢失。继续升温至 44℃和 46℃培养突变株BMB1 6 0和BMB1 70 ,其质粒可进一步丢失。在46℃培养条件下添加 0 .0 5 %SDS进行处理后 ,突变株BMB1 70质粒丢失的百分率提高 。 Plasmid stability of Bacillus thuringiensis influenced by treating it with ethidium bromide (EB), elevating growth temperature as well as adding sodium docecyl sulphate (SDS) were studied in this paper. Results showed that in wild type strains YBT?1520 and HD?567, the influence on stability of the plasmids carrying ICPs genes was not found as treating them with 4.5 μg/ml EB, whereas in wild type strains YBT?1463 and YBT?9603, there was 0.41 % and 0.37 % plasmids carrying ICPs genes as well as all small plasmids (less than 2 kb in size) lost, respectively, when their temperature reached 42℃. The plasmids of acrystalliferous mutant strains BMB160 and BMB170 further lost when elevating their growth temperature to 44℃and 46℃. The plasmid lost in mutant strain BMB170 increased, when adding 0.05% SDS to its medium at 46℃, and all plasmids in one colony even completely lost with this treatment.\;
