摘要
目的探讨黄芪总苷(TA)单独或与顺铂(DDP)联合应用对人子宫内膜癌KLE细胞生长的影响。方法培养KLE细胞,分为三组分别加入不同浓度TA和(或)DDP,另设阴性对照组,观察细胞形态学改变,中性红比色法检测细胞增殖,流式细胞仪检测细胞凋亡及周期分布。结果 TA能明显抑制KLE细胞增殖,且呈剂量-时间依赖性,12h、24h、48h的IC50分别为374.2、263.5、212.1μg/ml;两药联合应用时抑制率显著增高(P<0.01);流式细胞仪检测显示TA能使KLE细胞发生G1期阻滞,同时诱导细胞凋亡,其凋亡率与浓度-时间呈依赖关系,荧光显微镜可见细胞凋亡形态学变化。结论 TA可抑制KLE细胞增殖,诱导细胞凋亡,且与顺铂联合应用有协同作用。
Objective To study total astragalosides（TA）alone or in combination with cisplatin affect on the growth of endomet：nal cancer cell line KLE. Methods KLE cells were treated by TA at different concentrations. Cell growth and proliferation were observed by neutral red assay. Morphological changes of ceils were observed. Apoptosis and cell-DNA distribution was analyszed by flow cytometry. Results TA could inhibit KLE cell＇s proliferation. KLE cells viability in presence of TA decreased markedly in a dose-and time-dependent manner, IC50 was 374. 2,263.5, 212. 1 μg/ml at 12 hours,24 hours and 48 hours respectively, and the joint effect of the two drugs had significantly higher inhibition rate（P 〈0.01 ） ,showing their synergistic effects. Flow cytometry revealed that TA could cause G1 phase arrest and induce apoptosis at the same time; the apopiosis rate increased with the increase in drug concentration and the time. Conclusions TA can inhibit the proliferation of KLE ceils effectively and induce apoptosis,and the two drugs administered in combination show synergistic effects.
