摘要
采用高效液相色谱(二极管阵列)/电喷雾质谱联用系统(HPLC-PDA-ESI-MSn)快速鉴定中药粉葛中C-苷及O-苷异黄酮同分异构体。选用Hypersil Gold C18(150mm×2.1mm×3"m)色谱柱,以水(含0.1%甲酸)-乙腈为流动相进行梯度洗脱,研究了粉葛中11种异黄酮类化合物的正离子一级、多级质谱及PDA光谱;分析鉴别了两对同分异构体;预测了两种未知微量成分结构,建立了区分粉葛中异黄酮类化合物同分异构体的电喷雾质谱新方法。
C-glycosides and their isomer O-glycosides of isoflavones in radix puerariae thomsonii were rapid identified by high performance liquid chromatography photo diode array detection electrospray ionization tandem mass spectrometry (HPLC-PDA-ESI/MSn). The separation conditions employed for HPLC were optimized. A Hypersil Gold C18 column (3 μm, 2.1 mm×150 mm) was used in this study. The mobile phase consisted of water (0.1% formic acid) (A) and acetonitrile (B) in a gradient mode. The flow rate was 0.2 mL/min. Eleven isoflavones compounds included two pairs of isomers in radix puerariae thomsonii were identified by online photodiode array detection-MSn and the data obtained by this method were compared with that from the reported literature. Two unknown trace isoflavones isomers were predicted. This method could be used to quickly and conveniently distinguish the isomers isoflavones in Radix puerariae thomsonii.
