摘要
目的研究(热体克蛋白70)HSP70在对乙酰氨基酚诱导的小鼠急性肝损伤中的动态变化,进一步阐明HSP70在药物性肝损伤中的重要作用。方法健康雄性小鼠40只,分别腹腔注射对乙酰氨基酚溶液(用生理盐水稀释,550 mg/kg),在注射对乙酰氨基酚后62,44,2,54 h分别眼球取血,分离血清检测AST和ALT活性,同时取小鼠肝脏制备10%肝匀浆,每个时间点10只小鼠。同时取10只正常小鼠的血清和肝脏作为对照进行研究。利用蛋白印迹方法检测小鼠急性肝损伤过程中HSP70的表达变化。结果小鼠注射对乙酰氨基酚后,血清中AST和ALT活性随着时间的延长,酶活性逐渐升高,24 h达到高峰,54 h逐渐恢复到正常水平。HSP70在注射对乙酰氨基酚后6 h,其表达量略有增高,在24 h表达量达到高峰,说明小鼠肝脏的应激反应达到了高峰,此后HSP70的表达量逐渐恢复到正常水平。结论 HSP70在对乙酰氨基酚诱导的小鼠急性肝损伤过程中表达出现显著变化,可能在肝损伤的修复和再生过程中起到重要的作用。
Objective To study the dynamic changes of HSP70 during acute liver injury in mice induced by acetaminophen and further clarify the important roles of HSP70 in the drug-induced liver injury. Methods 40 healthy male mice were injected intraperitoneally with acetaminophen solution(diluted with saline,550 mg/kg).Eye blood was respectively collected at 6,24,42,54 h after acetaminophen injection.Serum was separated,then AST and ALT activity were measured.At the same time,the livers of mice were prepared for 10% liver homogenate.10 mice were used at each time point.At the same time,10 normal mice were used as controls.Western blotting was used to detect the expression changes of HSP70 during acute liver injury in mice. Results AST and ALT activities were gradually increased with time delayed and got to the peak levels at 24 h,then gradually recovered to the normal level at 54 h after acetaminophen injection.The expression of HSP70 was slightly increased at 6h and got to the peak levels at 24 h,which indicated that the emergency response of the mice liver got to the peak level.Then the amount of HSP70 expression gradually returned to normal levels. Conclusion The expression of HSP70 was significantly changed during acute liver injury in mice induced by acetaminophen,which may play an important role in the process of repair and regeneration of liver after liver injury.
