摘要
为了探讨柠檬酸对机体的毒副作用,选用昆明系健康雄性小鼠40只,随机分成4组,每组10只,饲养1周后,腹腔注射一定剂量的生理盐水及低(180 mg.kg-1)、中(270 mg.kg-1)、高(360 mg.kg-1)质量浓度的柠檬酸溶液.每周注射1次,连续注射3周,第3次注射后的第7天,所有小鼠均颈椎脱臼处死,采取小鼠的脑组织分别于液氮和-20℃各保存1份备用.通过RT-PCR法和ELISA法分别检测小鼠脑组织中bcl-2基因表达水平和蛋白表达量.结果显示,柠檬酸处理组与对照组相比,bcl-2基因表达水平均无统计学上的差异(P>0.05);中剂量柠檬酸处理时,bcl-2蛋白的表达量达到最高值,且与对照组呈现显著差异(P<0.05),上升了23%,柠檬酸高剂量组与中剂量组相比,bcl-2蛋白的表达量呈显著降低(P<0.05);其它组间均无统计学上的差异(P>0.05).说明腹腔注射270 mg.kg-1的柠檬酸可显著提高小鼠脑组织bcl-2蛋白的表达量;但本试验设置的柠檬酸梯度对bcl-2基因表达水平无显著影响.
To investigate the toxicity and side-effect of citric acid (CA) on the organic bady, 40 healthy Kunming mice were randomly divided into four groups, the control group and low, middle, high CA groups, treated with intraperitoneal injection of physiological saline and CA (180, 270, 360 mg·kg-1 ) respectively. Once a week with 0.2 mL injected, for three weeks, one week after the last treatment, all mice were killed by cervical dislocation, their brain was taken divided into two portions, and kept in liquid nitrogen and -20℃ refrigerator, respectively. The expression level of bcl-2 gene and the concentration of bcl-2 protein with were determined with RT-PCR and ELISA in the brain of the mice respectively. The results showed that there was no statistical difference (P 〉 0.05) between the CA treatment group and control group. The expression of bcl-2 protein increased along with CA concen- tration, up to peak at the moderate dose, and there was a significant difference between the moderate dose and the control group (P 〈 0.05) , with an increase of 23%. The high dose group significantly decreased compared with the moderate dose group (P 〈 0.05) , and there was no statistical difference in other groups (P 〉 0.05 ), which indicated peritoneal injection of 270 mg·kg-1 CA could significantly increas the bcl-2 protein expression in the brain of mice, but the concentration of CA in the experiment didn' t have significant effect on the gene expression of bcl-2.
