摘要
目的 探讨诊断超声对胎鼠中枢神经系统是否有刺激。方法 用诊断超声 (探头频率 5 .0MHz ,超声输出功率 1.1mW ,空间平均与时间平均声强 4.7mW /cm2 )辐照孕鼠腹部子宫体表投影区 ,分为照射 3 0min组、2 0min组、10min组和对照组。取胎鼠脑应用免疫组化法观察大脑中c fos基因表达的激活产物FOS蛋白标记的神经元 ,采用方差分析比较各组间差异。结果 3 0min组胎鼠脑海马区可见有群集性分布 ,着色深的FOS蛋白强标记神经细胞 ;2 0min、10min两组有散在分布 ,着色淡的FOS蛋白弱标记神经细胞 ;对照组未观察到FOS标记神经细胞。 3 0min组与其他各组均有显著性差异 ( P <0 .0 0 1) ,而2 0min、10min组和对照组间差异无显著性 (P >0 .0 5 )。结论 诊断超声辐照孕鼠达一定时间时 ,可致胎鼠脑海马区神经元c fos基因激活 ,其表达产物FOS蛋白增加 ,表明可以构成对胎鼠中枢神经元的刺激。
Objective To evaluate whether diagnostic ultrasound could stimulate fetal central nervous system of rats.Methods The diagnostic ultrasound ( 5.0 MHz) was used to irradicatie pregnant wistar rats,which were divided into 30 min,20 min,10 min and control groups.The fetal brain tissues were collected.The FOS postive neurons were observed by using immunohischemistry and the analysis of variance was used to compare the difference groups.Results There was in a crowd of FOS postive neurons in the fetal brain hippocampus in the 30 min group and scattered poor FOS postive neurons in the 20 min and 10 min groups.No FOS postive neuron was found in the control group.There were significantly differences (P< 0.001 ) between 30 min and other groups,but there was no significantly difference between 20 min and 10 min,20 min and control,10 min and control (P> 0.05 ).Conclusions When the pregnant rats were irradiated for 30 minutes,the expression of FOS protein and c fos gene expressing substance in felal hippocampus neurons was increased indicating that the fetal rats central neurons might be stimulated.
