摘要
目的:建立黄牡丹中芍药苷的含量测定方法并考察黄牡丹不同部位中芍药苷的含量。方法:采用高效液相色谱法,Agilent TC-C18色谱柱(4.6 mm×150 mm,5μm),流动相乙腈-0.1%磷酸溶液(12∶88),检测波长235 nm,流速1.0 mL·min-1,柱温30℃。结果:芍药苷进样量在0.40~4.00μg,与峰面积具有良好的线性关系,回归方程A=1 095.3 C+31.608(r=0.999 9),平均回收率100.59%,RSD 1.66%。芍药苷在不同部位中的含量分布为果实>叶>栓皮>皮部>茎>木部。结论:本方法简便、快速、准确,重复性好,芍药苷可作为控制黄牡丹药材质量的一个指标成分。
Objective: To develop a HPLC method to determine paeoniflorin in Paeonia delavayi var.lutea,and to investigate content of paeoniflorin in different parts of P.delavayi var.lutea.Method: The analysis was carried out at 30 ℃ on a TC-C18 column(4.6 mm×150 mm,5 μm) eluted with a mobile phase consisting of a mixture of acetonitrile-0.1% phosphoric acid(12∶88).The flow rate was 1.0 mL·min-1;and the detected wavelength was 235 nm.Result: The calibration curve was linear within the concentration ranges of 0.40-4.00 μg.The regression equation was A=1 095.3 C+31.608(r=0.999 9).The average recovery was 100.59%,while RSD was 1.66%.The concentration distribution of paeoniflorin in different parts of P.delavayi var.lutea is as follows: fruitleafcorkcortexstemxylem.Conclusion: This method is simple,accurate and with good repeatability.The paeoniflorin can be selected as a quality control component in P.delavayi var.lutea.
