摘要
目的 :观察高压氧与甲基强地松龙对不同系小鼠皮肤移植局部浸润 T淋巴细胞亚群及细胞粘附分子表达的影响。方法 :C5 7BL/ 6小鼠接受 BAL B/ C小鼠皮肤移植后 ,分别在 99.2 % O2 、0 .2 5 MPa高压氧舱内作用 1.5h/ d,并每日腹腔注射 0 .17%甲基强地松龙 0 .3m l,14d后利用免疫荧光染色技术和图像分析仪测定 CD3、CD4、CD8、CD1 1 a、CD1 8阳性细胞荧光面积。结果 :高压氧组、甲基强地松龙组、高压氧与甲基强地松龙联合组可使局部的组织破坏减轻 ,使 CD3、CD4、CD8、CD1 1 a、CD1 8阳性细胞荧光面积明显低于对照组。其中 ,高压氧组优于甲基强地松龙组 ,高压氧与甲基强地松龙联合组明显优于单纯应用高压氧组或甲基强地松龙组 (P <0 .0 1)。结论 :高压氧与甲基强地松龙可减少移植局部 T淋巴细胞及其亚群的浸润及粘附分子的表达 ,从而抑制了移植排斥反应 ,且两者联合应用具有协同作用。
Objective:We observed the effect of hyperbaric oxygen(HBO) and methylpred disolonum(MD) in local lymphocyte infiltration and expression of adhesion molecules in the heterodermic graft in mice. Methods:Skin transplantation, donor mice BALB/C, receptor mice C 57 BL/6 HBO group were treated by 99.2% oxygen with 0.25 MPa for 1.5 hours every day. MD group were treated by MD 25 mg/kg·d -1 by abdomen injection. HBO+MD group were treated with both ways at the two groups. Control group were treated by 0.9% N S.After 14 days,T lymphocyte and CD 11a 、CD 18 in local graft were detected with immunofluoresence techniques and image processing system. Results:All indexes of HBO group, MD group and HBO+MD group were lower than those of control group(P<0.01),the indexes of HBO+MD group are the lowest. Conclusion:HBO and MD have inhibition effect on local lymphocyte infiltration and the expression of adhesion molecules andhence toinhibitimmune rejection.
