摘要
利用犬瘟热病毒 (caninedistempervirus,CDV)弱毒标准株OP CDV感染敏感细胞系非洲绿猴肾细胞 (Vero) ,应用缺口末端标记法 (TdT mediateddUTPnickendlabeling ,TUNEL)分析感染细胞的凋亡。结果在感染后 48h的Vero细胞中检出了凋亡阳性细胞 ,而对照细胞内未检测到凋亡细胞。感染后 2 4h的细胞尚未出现细胞病变 ,这一时期的细胞内也未检测到凋亡细胞。表明CDV能诱导细胞调亡 ,且凋亡细胞的检出时间与细胞病变的出现时间呈正向相关。
TdT mediated dUTP nick end labeling(TUNEL)method was used to detect apoptotic cells in Vero cells infected by canine distemper virus(CDV).Control cells included uninfected cells treated by nomal procedure of the detection kit and infected cells at 48 h postinfection(p i.)treated with reagents excluded terminal deoxynucleotidy 1 transferase(TdT).The apoptotic cells were only obeserved in infected Vero cells at 48 h p i.,while control cells did not show apoptosis.In the meanwhile,the infected cells underwent gradually typical cytopathic effect(cpe)with the time progress.At 24 h p i.,cpe was not developed yet in infected cells.But at 48 h p i.,the infected cells had already experienced apparent cpe.Coincidentally,the apoptotic cells in infected cells could not be detected at 24 h p i.but could be detected at 48 h p i..It concluded that CDV could induce apoptosis of infected Vero cells.Futhermore,apoptosis in infected Vero cells had a similar time course to cpe.
