摘要
目的观察泽泻含药血清对H2O2诱导损伤的血管内皮细胞活性、形态学、SOD及NO的影响,探讨泽泻含药血清抗氧化损伤的可能机理。方法 100μmol/L H2O2造成血管内皮细胞氧化应激损伤模型,cck-8法检测各组细胞增殖情况;黄嘌呤氧化酶法和硝酸还原酶法检测各组细胞上清液中SOD的活力和NO含量。结果 100μmol/L H2O2可抑制血管内皮细胞活性,降低SOD活力和NO的分泌,而泽泻则能改善100μmol/L H2O2对血管内皮细胞的损伤,对血管内皮细胞具有一定的保护作用。结论泽泻可以通过调控抗氧化损伤机制保护血管内皮细胞,这可能是泽泻抗氧化损伤机理所在。
OBJECTIVE To observe the influence of the serum containing active ingredients of Alismatis Rhizoma on viability,morphology,SOD,and NO of vascular endothelial cells damaged by H2O2,and to study the possible mechanism of anti-oxidative damage of the serum on the cells.METHODS The endothelial cells treated by 100 μmol/L H2O2 were used as an oxidative stress model.The method of cck-8 was applied to detect cell proliferation;xanthine oxidase and nitrate reductase assay were utilized to detect SOD activity and NO content in the medium supernatant of each group,respectively.RESULTS In model groups,viability of vascular endothelial cells was inhibited partly,while their SOD activity and NO secretion decreased.But,in administration groups the oxidative damage of vascular endothelial cells was reduced and the active ingredients of Alismatis Rhizoma had a protective effect on oxidatively damaged vascular endothelial cells.CONCLUSION The mechanism of oxidative damage of the active ingredients of Alismatis Rhizoma may be related to its reduction of the oxidative damage of endothelial cells to improve the cell survival.
