摘要
目的:探讨环氧合酶-2(COX-2)和基质金属蛋白酶-2(MMP-2)在结肠癌组织中的表达并讨论其临床意义。方法:采用免疫组织化学SP法检测100例结肠癌组织和40例癌旁结肠正常组织中COX2、MMP-2的表达情况。结果:在结肠正常组织和结肠癌中,COX-2蛋白的阳性表达分别为7.5%(3/40)和83.0%(83/100),而MMP-2蛋白阳性表达分别为5.0%(2/40)和94.0%(94/100),COX-2和MMP-2在结肠癌组织中的阳性表达均明显高于结肠正常组织(P<0.01)。在结肠癌组织中,COX-2和MMP-2阳性表达临床中晚期高于早期,有淋巴结转移者高于无转移者(P<0.05)。COX-2和MMP-2两者阳性表达呈正相关(r=0.365,P<0.05)。结论:结肠癌中COX-2和MMP-2阳性表达明显升高,且与临床分期和淋巴结转移相关。检测COX-2和MMP-2表达有助于判断结肠癌的潜在转移和预后分析。
Objective： To detect the expression of cyclooxygenase-2 （COX-2） and matrix metalloproteinase-2 （MMP-2） in invasive breast carcinoma, and explore their correlation. Methods： The expression of COX-2 and MMP-2 were tested by SP immunohistochemistry in 100 cases of colon cancer and 40 cases of normal colon tissues, Results：The positive expression of COX-2 protein in normal colon tissue and colon cancer were 7.5% （3/40） and 83.0% （83/100）, in MMP-2 they were 5.0% （2/40） and 94.0% （94/ 100） respectively, and expression positive rates of COX-2 and MMP-2 in colon cancer tissue were significantly higher than normal colonic tissue （P〈0.01）. The positive expression COX-2 and MMP-2 in the colon cancer, clinical middle-late stage was higher than the early, and lymph node metastasis was higher than those without the transfer （P〈0.05）. Positive correlation could be found between the expression of COX-2 and that of MMP-2 as well （r=0. a65,P〈 0.05）. Conclusions：COX-2 and MMP-2 protein are highly expressed in colon cancer and are all related with clinical stages and lymph node metastasis. These proteins are indicators of metastasis and predictors for prognosis of colon cancer.
