摘要
为缩短紫斑牡丹育种周期,建立牡丹胚培养研究的最佳体系,通过培养基的筛选,选择不同采收期的种子通过不同外植体类型进行紫斑牡丹胚培养试验。结果表明:紫斑牡丹胚培养最适启动培养基为1/2 MS+1.0 mg/L IAA+1.0 mg/L GA3,最适根诱导培养基为1/2 MS+0.05%AC+0.1 mg/L IBA,最适继代生长培养基为1/2 MS+0.05%AC;种子成熟度的不同会影响幼苗的伸长生长;外植体不同类型会影响种子的萌发情况。
In order to shorten the breeding cycle of Paeonia rockii and to provide the best system for embryo-culture,different seed collection time of seeds were selected through culture media selection and different explant types were chosen to conduct an embryo-culture experiment for Paeonia rockii.The results showed that the optimal initial medium for Paeonia rockii is 1/2 MS+1.0 mg/L IAA+1.0 mg/L GA3,the optimal rooting medium is 1/2 MS+0.05% AC+0.1 mg/L IBA,the optimal offspring medium is 1/2 MS+0.05%AC;different maturities of seed will influence elongation growth of seedlings;different explant types will impact seed germination.
