摘要
目的探讨碱性成纤维细胞生长因子(bFGF)和转化生长因子-β(1TGF-β1)单独及联合应用时对人牙龈成纤维细胞(HGF)增殖及I型胶原的分泌和糖胺多糖(glycosaminoglycan,GAG)合成的影响。方法体外培养HGF,MTT法动态检测两种生长因子单独及联合作用后的增殖效应;羟脯氨酸试剂盒消化法及阿利新蓝法分别检测最佳增殖浓度时上清液中I型胶原及糖胺多糖含量。结果第3天开始bFGF和TGF-β1促进HGF增殖,第5天最显著(P<0.05),bFGF和TGF-β1的浓度分别为10 ng/mL和1 ng/mL时增殖效应最佳,联合作用更显著(P<0.05);最佳增殖浓度时TGF-β1促进I型胶原分泌,而bFGF起抑制作用(P<0.05),联合应用时TGF-β1的促进作用占优势,但作用减弱(P<0.05);最佳增殖浓度时bFGF和TGF-β1促进GAG合成(P<0.05),联合应用时更显著(P<0.05)。结论除bFGF单独作用时抑制I型胶原的合成外,bFGF和TGF-β1单独及联合应用时能促进HGF增殖和细胞外基质的合成。
【Objectives】 To study the effects of TGF-β1 and bFGF alone and combined effects on proliferation of human gingival fibroblast cells as well as the effects on extracellular matrix type I collagen and GAG content,and to choose the best markedly preferred concentration.【Methods】 TGF-β1 or bFGF of different concentration were added into DMEM culture individually or combined.HGF proliferation was measured with MTT colorimetric assay.Hydroxyproline kit digestion and the dynamic effects of different concentrations of bFGF and TGF-β1,respectively,on separate and combined effects on type I collagen in the supernatant of GFCs content.Alcian blue method of dynamic determination of bFGF and TGF-β1 concentration gradient medium GAG content.【Results】 TGF-β1 and bFGF could increase the HGF proliferation as well as the content of GAG alone and combined.The effect on proliferation and the content of GAG in combined group is significantly better than in the individual groups(P 〈0.05).The best markedly preferred concentration is 10 ng/mL(bFGF) and 5 ng/mL(TGF-β1).TGF-β1 increased the content of extracellular matrix type I collagen,whereas bFGF had opposite effects.When both growth factors were associated,the anabolic effects observed of TGF-β1 treatment seem to predominate,however attenuated.【Conclusion】 TGF-β1 and bFGF could increase the HGF proliferation and the content of the extracellular matrix type I collagen and GAG combined,and provide partly theoretical basis in gingival regeneration.
