摘要
目的观察荞麦黄酮复方制剂(FBC)对糖尿病大鼠肾脏损伤的抑制作用并探讨其可能的机制。方法雄性SD大鼠用高脂饲料和STZ建立大鼠2型糖尿病模型,随机分为模型对照组(蒸馏水10 ml.kg-1.d-1),阳性药物组(消渴丸0.83 g.kg-1),FBC低、高剂量组(FBC 0.6,1.2 g.kg-1);另取正常鼠为正常对照组(蒸馏水10 ml.kg-1.d-1),各组大鼠灌胃给药连续8周。观察大鼠一般状况,用血糖仪测空腹血糖(FBG)、24 h尿蛋白定量、血和尿肌酐、内生肌酐清除率、双肾指数、血和肾组织内皮素(ET-1)及糖基化终产物(AGEs)、肾组织病理改变。结果与正常对照组比:模型组FBG明显升高,肾重指数和24h尿蛋白排泄量明显升高,而内生肌酐清除率却明显降低,血和肾组织ET-1、AGEs升高及肾组织形态结构有明显改变;高、低剂量FBC能不同程度的改善上述指标的变化,明显降低血糖,增加肾肌酐清除率,肾组织ET-1、AGEs含量明显低于模型组,FBC可使肾损伤明显减轻,且高剂量作用更明显,与消渴丸组相近。结论 FBC对STZ所致糖尿病大鼠肾损伤有明显的抑制作用,其机制可能是通过降低血糖,抑制AGEs、ET-1形成从而减少肾损伤。
Objective To observe the inhibitory effect and explore its mechanism of FBC on kidney injury of diabetic rats. Meth- ods The type 2 diabetic male SD rats model was induced by feeding high - fat diet and intraperitoneal injection with streptozotoein rats. The rats were divided into five groups randomly:model group( distilled water, lOml ~ kg-1 , d- 1 ) , positive medicine groups (XKW, 0.83g ~ kg-l . d-l) , the low and high dose FBC group (FBC ,0.6 g · kg^-1· d^-1 and 1.2 g · kg^-1·d^-1) ,and control group( distilled water, 10 ml · kg^-1· d^-1 ). Each rat was garaged daily for eight weeks. The daily condition of rats, level of FBC, 24h urine protein, level of cr in blood and urine, Ccr, index of kidney, ET - 1 and ACEs in blood and kidney, and pathological changes in kidney tissue were observed respectively. Results Compared with normal control group, the level of FBG, renal index, and 24h urine protein in model group were significantly higher;and the level of Ccr decreased significantly. ET - 1 and AGEs of blood and kidney increased, and renal morphological changed significantly. Compared with model group, the level of FBC could be improved. The changes in above indicators:the level of FBG, ET - 1 and AGEs of blood and kidney decreased significantly, the level of Ccr increased, and FBC could reduce renal injury significantly. The higher level of FBC, the more obvious effects, similar- ly with XKW group. Conclusion FBC can inhibit renal injury of diabetic rats induced by STZ obviously, and the mechanism may reduce the renal injury by lowering blood sugar, inhibiting the formation of ET - 1 and ACEs.
