摘要
目的建立测定天宁颗粒中阿魏酸的高效液相色谱(HPLC)法。方法采用反相高效液相色谱法测定阿魏酸的含量。应用Diamonsil ODS C18色谱柱(200 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(20∶80),检测波长为322 nm,流速:1 mL.min-1;进样量:20μL;柱温:室温。结果阿魏酸浓度在1.0~100.0μg.mL-1范围内线性关系良好,r=0.999 6;平均回收率为99.78%,RSD=2.96%(n=9)。结论该方法专属性强,操作简单,结果准确,重复性好,可用于天宁颗粒中阿魏酸的含量测定。
Objective To establish a HPLC method for determination of ferulic acid in tianning granules. Methods The RP-HPLC method was used to determine the content of ferulic acid. The separation was achieved on Diamonsil ODS Cl8 column(200 min×4. 6 mm,5 μm)with a mobile phase of acetonitrile-0. 1% phosphoric acid (20 : 80) at a flow rate of 1 mL · min^-1 by loading with 20μL of samples, the wave length was set at 322 nm and the process was carried out at room temperature. Results The linearity range of ferulic acid was within 1.0-100.0μg·mL^-1, r = 0. 999 6. The average recovery was 99.78% , RSD = 2.96% ( n = 9 ). Conclusion The HPLC method is simple, accurate, reproducible and highly selective, which can be used to determine the content of ferulic acid in tianning granules.
