犀角地黄汤合银翘散对流感病毒性肺炎小鼠炎性因子和自由基表达的影响 被引量:11

Effect of Xijiaodihuangtang and Yinqiaosan on Inflammatory Cytokines and Free Radicals in Mice with Pneumonia Caused by Influenza Virus

摘要 目的观察犀角地黄汤合银翘散对流感病毒感染致病毒性肺炎小鼠炎性因子、自由基表达的影响。方法Balb/c小鼠108只随机分为正常组、模型组、药物组,25μL 50LD50病毒液滴鼻建立流感病毒鼠肺适应株感染的小鼠肺炎模型(除正常组外),感染后1 h,正常组和模型组予以双蒸水灌胃,药物组予中药23 g/(kg.d)灌胃,各组均给药2次/d,连续5 d。感染后2、4、6 d分别取材,采用酶联免疫吸附法检测血清中肿瘤坏死因子-α(TNF-α)、肺组织匀浆中单核细胞趋化蛋白-1(MCP-1)的含量,生化法检测肺组织中超氧化物歧化酶(SOD)、一氧化氮(NO)、一氧化氮合酶(iNOS)、丙二醛(MDA)含量。结果临床等效剂量的犀角地黄汤合银翘散在感染后第4、6日可明显降低TNF-α的含量(P<0.01,P<0.05);感染后第6日,抑制MCP-1的表达(P<0.01),升高SOD活性(P<0.01);在各时间点均降低MDA含量(P<0.01);感染后第2日,明显降低iNOS的含量(P<0.01),第4、6日,明显降低NO含量(P<0.01)。结论犀角地黄汤合银翘散可明显抑制流感病毒致病毒性肺炎小鼠体内炎性细胞因子及氧自由基的产生,具有抗炎作用。 Objective To explore the effect of classical combined formula Xijiaodihuangtang and Yinqiaosan on inflammatory cytokines and free radicals in mice with viral pneumonia caused by influenza virus.Methods A total of 108 Balb/c mice were randomly divided into control group,model group,and Chinese medicine group.25 μL 50LD50 influenza virus,mouse lung-adapted strain,was intranasally inoculated to mice except the control group.1 h after infection,mice in control and model group were intragastrically given 25 μL distilled water by day 5,mice in Chinese medicine group were intragastrically given Chinese medicine at a dose of 23 g/(kg?d) by day 5.On day 2,4 and 6 after infection,ELISA method was used to detect the concentration of tumor necrosis factor-alpha(TNF-α) in serum and monocyte chemoattractant protein-1(MCP)-1 in lung,biochemical detection was used to measure the concentrations of SOD,MDA,NO and iNOS in lung tissue.Results The clinical equivalent dose of Xijiaodihuangtang and Yinqiaosan obviously reduced the concentration of TNF-α on day 4 and 6(P〈0.01,P〈0.05),inhibited the expression of MCP-1(P〈0.01) and increased the activity of SOD on day 6 after infection(P〈0.01).Xijiaodihuangtang and Yinqiaosan reduced the concentration of MDA at each timepoint(P〈0.01),reduced the concentration of iNOS on day 2(P〈0.01) and the concentration of NO on day 4 and 6 after infection(P〈0.01).Conclusion Xijiaodihuangtang and Yinqiaosan obviously inhibited the production of inflammatory cytokines and free radical in mice with viral pneumonia caused by influenza virus.
