赤芍总苷对大鼠放射性肝纤维化的保护作用及机制 被引量:22

Protective effect and mechanism of total paeony glycoside on radiactived hepatic fibrosis in rats

摘要 目的:探讨赤芍总苷对大鼠放射性肝纤维化的保护作用及机制。方法:雄性大鼠放射性肝纤维化动物模型,将大鼠随机分为正常对照组、模型组、赤芍总苷组(50 mg/kg、25mg/kg),除正常对照组外,其余各组均制备成放射性肝纤维化模型,造模后各组每天给予相应药物灌胃。于照射后第26周末剖杀大鼠,取血和肝脏标本,ELISA法检测大鼠中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN含量,碱水法检测大鼠肝脏组织中Hyp含量;光镜下观察肝脏HE染色、MASSON染色病理改变;免疫组化法测肝脏组织TGF-β1、Smad3/4/7蛋白表达情况。结果:与正常对照组比较,模型组大鼠肝脏损伤和胶原纤维增生明显,血清中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN含量明显增加(P<0.01),肝脏组织中Hyp含量增加(P<0.01),肝脏组织中TGF-β1、Smad3/4/7蛋白表达增加(P<0.01)。与模型组比较,赤芍总苷治疗组(50mg/kg)可显著降低肝纤维化大鼠血清中TGF-β1、TNF-α、IL-6、HA、PC-Ⅲ、LN的含量,同时可降低肝脏组织中Hyp含量及降低肝脏损伤程度及胶原纤维增生程度,还可减少大鼠脏肝组织中TGF-β1和Smad3/4蛋白表达,增加肝脏组织中Smad7蛋白表达。结论:赤芍总苷具有明显的抗肝纤维化作用,其机制可能与其阻断TGF-β1/Smad信号传导通路有关。 To study the protection and mechanism of Total paeony glycoside (TPG) on radiactived hepatic fibrosis in rats. Methods: Establish experimental animals models of radiactived hepatic fibrosis in male SD rats. The rats were randomly divided into Normal control group, Model group, TPG group (50mg/kg). The effects of TPG were observed on the levels of serum TGF-βI ,TNF-α, IL-6, HA, PC-Ⅲ, LN and Hyp in rats liver fibrosis, and on liver histopathological changes as well as collagen hyperplasia formation in liver tissue. Liver samples were collected after experiments and stained by immuninochemistry of TGF-βI and Smad3, 4, 7. Results : TPG 50mg/kg can significantly reduce serum TGF-βI ,TNF-α, IL-6, HA, PC-Ⅲ, LN and Hyp contents in liver fibrosis of rats, improve the liver pathologic injury, reduce collagen hyperplasia in liver of liver fibrosis rats. TPG 50mg/kg treatment could significantly down-regulated TGF-β1 Smad3/4 expression and up-regulated Smad7 expression. Conclussions: TPG has the protective effect against radiactived hepatic fibrosis in rats. Its mechanism may be related to blocking TGF-β1/Smad signal transduction pathway.
